Dorsomorphin is a cell permeable pyrrazolopyrimidine compound that act as potent inhibitor of AMP-activated protein kinase (AMPK) (Ki=109nM) and bone morphogenic protein (BMP) signaling (1,2). Dorsomorphin inhibits BMP type I receptors ALK2, ALK3 and ALK6 and thus blocks BMP-mediated SMAD1/5/8 phosphorylation (3). It has also no effect on components of the ZAPK, Syk, PKCq, PKA, or JAK3. It is shown that dorsomorphin promotes hESC self-renewal by antagonizing autocrine BMP signaling (4).
